Barry Kripke stops by to thank Amy for helping him with her ideas on string theory, which bothers Sheldon because she never helped him out when he was working on it. He complains to Penny who tells him to "let it go". Instead, Sheldon reveals that Amy was secretly performing experiments on Penny and Leonard and comparing the results with those of a chimpanzee, infuriating Penny. Leonard and Raj run into Nathan Fillion at a deli, but he denies he is an actor so he can eat in peace. Later, he admits who he is and offers to take a picture with them. Raj asks so many questions to verify Fillion's identity that he looks annoyed in the photo. Stuart has opened his new comic book store, which impresses everyone except Howard who finds him using the den furniture that his mother had given him. However, all arguments end when Howard gets a call from Florida where his mother is visiting, and receives news that she died in her sleep. Everyone, including Sheldon, goes to comfort Howard. After he and Bernadette leave for Florida, the rest of the gang remember and toast the death of Mrs. Wolowitz, calling her "everyone's mother".

Beim gemeinsamen Mittagessen erfährt Sheldon, dass Amy Barry Kripke bei seinen Forschungen zur String-Theorie unterstützt hat, was ihm besonders dadurch bitter aufstößt, da er selbst die Forschung an der String-Theorie aufgegeben hatte, weil er sie als Sackgasse sah. Als Penny daraufhin versucht, Sheldon beizubringen, loszulassen, plaudert Sheldon aus, dass Amy in der letzten Zeit Penny immer wieder Rätselaufgaben gab, um ihre Intelligenz mit der ihrer Laboraffen zu vergleichen, woraufhin wiederum Penny wütend auf Amy wird und sie im Hausflur damit konfrontiert. Stuart feiert derweil die Wiedereröffnung seines Comicbuchladens, bei der Howard feststellen muss, dass Stuart geschenkte Möbel aus der Wohnung seiner Mutter im Laden aufgestellt hat. Diesen Umstand empfindet Howard im Anbetracht der Tatsache, dass Stuart es überhaupt erst Howards Mutter zu verdanken hat, dass er den Laden wiedereröffnen konnte, als unangemessen. Während Leonard und Raj beim Organisieren des Essens auf Nathan Fillion treffen, versucht Bernadette Howard aufzubauen, indem sie anmerkt, dass der Erfolg von Stuarts neu eröffnetem Laden ihm wieder auf eigene Beine helfen könnte, was Howard nicht hilft. Als sich alle Parteien im Comicbuchladen versammeln, erschüttert eine tragische Nachricht die Gruppe: Howards Mutter, welche in Florida bei seiner Tante Gladys zu Besuch war, ist gestorben.

Stuart vient d'ouvrir une nouvelle boutique de comics (vu que la dernière a pris feu). Ses amis viennent pour l'ouverture alors que de petites disputes commencent : Amy utilisait Leonard et Penny pour des expériences cognitives et Sheldon a découvert qu'Amy a aidé Barry Kripke dans ses recherches. Howard, quant à lui, apprend le décès de sa mère.

Sheldon scopre che Amy ha aiutato Kripke nelle sue ricerche sulla teoria delle stringhe e si arrabbia; parlandone con Penny, le rivela che Amy l'ha segretamente sottoposta ad alcuni test per vedere se fosse più intelligente delle scimmie che sta studiando. Nel frattempo Stuart riesce a riaprire il suo negozio di fumetti, ma quando Howard scopre che sua madre, oltre ad un importante aiuto economico e all'ospitalità che gli ha offerto, gli ha anche regalato alcuni mobili della casa si arrabbia. Alla fine Penny e Leonard, sottoposto da Sheldon a test simili a quelli della fidanzata, ottengono le scuse di Amy e del suo ragazzo, ma proprio in quel momento giunge una notizia terribile: la madre di Howard, in vacanza dalla sorella in Florida, è morta nel sonno. Howard e Bernadette partono subito per il funerale, mentre i loro amici e Stuart fanno un brindisi per la signora Wolowitz.

Howard: ...I'm sorry, but it's making me crazy. Bernadette: Can you please just let it go? Sheldon: Ooh, I can help you with that. Imagine you're holding an ordinary pen - while your favourite pen is safe and secure in your pocket. [Howard's phone rings] Howard: Hold that thought. [goes off to answer it in the next room] Hello? Amy: [To Leonard and Penny] How can I make this up to you? Penny: [angrily putting both fists together] The answer is in this puzzle box, let's see if you can open it! [She and Leonard walk away, while Bernadette goes to Stuart] Bernadette: You could have at least warned him about the furniture. Raj: That's what I said when we moved it! Bernadette: You helped him!? Raj: [sarcastically] No. Stuart picked up those three pillows all on his own. [Howard returns from his phone call, looking visibly shaken] Leonard: Hey, you okay? Howard: [in a dazed voice] No... Leonard: What's wrong? Howard: ...My mom died. [All heads turn towards him in shock] Bernadette: What!? Howard: That was my aunt. Ma took a nap and... She never woke up. Bernadette: [beginning to cry] Oh my God, Howie... [She embraces him, while the rest of the group solemnly gather round, Penny visibly looking on the verge of tears as well] Leonard: I'm so sorry. Raj: What can we do? Howard: I don't know... [He sits down in an armchair and buries his face in his hands] Sheldon: May I say something? Leonard: [sadly] Not right now, Sheldon. Sheldon: But I think it would be comforting. Leonard: Buddy - Howard: It's okay... [looks up at him] What? Sheldon: When I lost my own father, I didn't have any friends to help me through it. You do. [Howard gives him a weak smile, while the others remain in shocked silence, both from the news and the fact that Sheldon has said something genuinely supportive] Penny: [breaking down] I really thought he was gonna say "let it go."
