Leonard is jealous of Penny's British classmate Cole who works with her on an oral class project, afraid Cole is hitting on her. Confronting him on the staircase, he tries to frighten him off Penny, unaware he is uninterested. She overhears them and is furious at Leonard's lack of trust. In the Caltech cafeteria, Leonard discusses his problem with Alex, who flirts with him, unbeknown to him. He later tells Penny of his insecurity about other men hitting on her, and she finally confesses her love for him. Meanwhile, Howard and Raj wonder what Sheldon does from 2:45 to 3:05 pm daily. They see him enter a storage room in Caltech's basement. They break in at midnight to find only the number "43" on a chalkboard. To discover its meaning, Howard installs a Mars Rover camera to spy on Sheldon. Finding the camera, he fakes a video of himself in the room under attack by an alien creature after he opens a small wormhole to find life in the 44th parallel universe, the first "43" being lifeless. He admonishes them for spying and reveals he relaxes in the room from his difficulties in daily life, but will not explain "43". The audience see Sheldon playing with a hacky sack in the room: "43" is his maximum number of consecutive hits.

Howard: "Er hat über zwei Jahre für den Todesstern gebraucht, da mache ich mir keine Sorgen." ----------------- Penny zu Howard: "Hey, musst Du nicht mit Deiner Gang um die Häuser ziehen und Gleichungen an die Wände sprayen?
Howard bemerkt, dass Sheldon sich jeden Tag von 14:45 bis 15:05 von der Gruppe trennt, und nicht einmal seine Assistentin Alex weiß, was er in dieser Zeit tut. In der Folge wollen Rajesh und er unbedingt hinter das Geheimnis kommen, und folgen Sheldon heimlich zu einem kleinen Raum im Keller der Universität. In der folgenden Nacht brechen beide dort ein, finden aber nur einen unscheinbaren Raum mit einer Tafel vor, auf der die Zahl 43 geschrieben steht. In der Folge zerbrechen sie sich ergebnislos den Kopf darüber, was es mit der Zahl auf sich haben könnte. Schließlich installiert Howard eine Kamera, um Sheldon dort beobachten zu können. Dieser entdeckt die Kamera jedoch und spielt den beiden einen üblen Streich, so dass am Ende nur dem Zuschauer das eher banale Geheimnis des Raumes offenbart wird. Leonard ist derweil eifersüchtig auf Pennys neuen Mitschüler, da er gut aussieht und einen britischen Akzent hat. Auch die Gespräche darüber mit Sheldon (dessen Ausführungen zu dem Thema Leonard eher noch mehr beunruhigen) und Alex (die mit Leonard flirtet ohne dass dieser es bemerkt) können nicht verhindern, dass er sich zu einer peinlichen Aktion gegenüber dem vermeintlichen Nebenbuhler hinreißen lässt. So kommt es zwischen ihm und Penny zum Streit, in deren Verlauf sie ihm das erste Mal überhaupt sagt, dass sie ihn liebt..

Howard se da cuenta de que Sheldon está desapareciendo durante veinte minutos cada tarde y que nadie sabe a dónde va ni por qué. A raíz de Sheldon, Howard y Raj saben que él va hacia el sótano, donde se encuentran con una pizarra con el número "43" escrito en él. Por otra parte, Leonard se pone celoso cuando se entera de que Penny supone que debe hacer una asignación con un estudiante británico llamado Cole. Él habla con Alex sobre el problema, pero no se da cuenta de que ella está interesada en él, entristeciendo a Alex.
Howard y Raj ven el video al día siguiente (con la cámara de repuesto de un vehículo en Marte que Howard robó para el evento) y ven a Sheldon ir y abrir un agujero de gusano pequeño con su "generador de agujeros de gusano". Él menciona que no ha encontrado vida en 43 Universos paralelos y que tratará de encontrar en el universo número 44, mete la cabeza por el agujero de gusano y sale gritando con una criatura atacando a su cara. Raj y Howard ven cómo Sheldon grita, emerge desde atrás y deja caer la criatura (de plástico) en su computadora portátil. Sheldon había encontrado su cámara oculta, y generó el clip de "retocada" del vídeo a manera de venganza. Sheldon les regaña, le explica que necesita 20 minutos para sí mismo y los deja sin revelar su secreto. Mientras tanto, en el sótano, Sheldon está secretamente tratando de rebotar un Hacky Sack 43 veces, su mejor marca personal, todos los días. Él falla esta vez.
Mientras tanto, Leonard intenta hablar con Penny y más tarde confiesa que no le gusta ser golpeado todo el tiempo. Penny le dice que ella eligió para que estuvieran con él y que ella lo ama. Al darse cuenta de que ella le confesó su amor hacia él, por primera vez, el dúo está de acuerdo en no decir nada a nadie.

Leonard est persuadé qu'il en profite pour draguer Penny et s'inquiète donc pour le futur de sa relation. Pendant ce temps, Howard remarque que Sheldon disparaît mystérieusement 20 minutes tous les jours et avec Raj, ils vont tenter d'en découvrir la raison.

Leonard è geloso di Cole, il nuovo compagno di studi di Penny, ma la ragazza lo rassicura dicendogli che sono solo amici e, mentre lo fa, gli dice per la prima volta che lo ama. Intanto Raj e Howard cercano di scoprire dove sparisce tutti i giorni Sheldon dalle 14.45 alle 15:05, perciò lo seguono fino ad un vecchio ripostiglio dell'università dove trovano scritto su una lavagna il numero 43. Non riuscendo a capire cosa significhi, i due mettono una telecamera nel ripostiglio e vedono Sheldon che fa dei test con un generatore di wormhole che ha creato, il quale gli permette di creare portali per universi paralleli. 43 sono i test che ha fatto e gli universi paralleli che ha controllato alla ricerca di vita; nell'esperimento numero 44, che i due vedono in diretta, trova qualcosa e precisamente un alieno che lo attacca. Raj e Howard vengono spaventati a morte da quanto vedono, fino a che Sheldon rivela loro che ha manomesso il collegamento video facendo vedere una cosa falsa, aggiungendo che si chiude nel ripostiglio perché ha bisogno di 20 minuti al giorno per spegnere la mente e ricaricarsi e che non rivelerà mai loro cosa fa lì e cosa significhi 43. Agli spettatori però il mistero viene svelato alla fine dell'episodio: Sheldon si reca in quel ripostiglio a palleggiare con una semplice pallina rimbalzante, e 43 non è altro che il suo record.
